The high school boy Miles (voiced by Shameik Moore) is unexpectedly bitten by a spider that has been exposed to radioactive contamination, leading to strange changes in his body. By chance, he witnesses a battle between Spider-Man (voiced by Chris Pine) and the crime boss Kingpin (voiced by Liev Schreiber). To prevent Kingpin from using a collider to disrupt the universe, Spider-Man ultimately sacrifices himself heroically. Not long after, the despondent Peter Parker from a parallel universe (voiced by Jake M. Johnson) appears in front of Miles. To stop Kingpin's conspiracy and help this Parker return to his own universe, they break into Kingpin's lair. In the process, they are attacked by Doctor Octopus and the Prowler.
